I'm really unhappy about not using Mother, as I didn't exactly see her as an antagonist, however she does work.

The final boss fight has it's own theme, which counts correct?

Yeah she even has two themes during her fight. You could even say all the themes playing in the ruins count since she is the ruins.

I would kinda question whether or not she's the "main" antagonist since League's lore is all over the place and there's no single evil force, but I do suppose she's the mastermind behind some of the more recent lore developments and might be the biggest threat to the world of League right now maybe??

She's trying to bring back the watchers right? Wouldn't they be higher up the villain list?
Not to mention all the other evil champions that want everyone dead

She seems to be a main antagonist, so I'll count it.
